Developing microscopy based microfluidic SLS assay for on-chip hemoglobin estimation

Point-of-care (POC) biochemical assay is a highly important to estimate hemoglobin in the blood. High reagent volumes and complex-expensive optical setup requirements pose serious challenges when it comes adopting conventional assays such as Sodium Lauryl Sulfate (SLS) method into POC device. Here, we report modified SLS on microfluidic platform, wherein quantification achieved using simple microscopy-based imaging setup. Assay parameters, including reagent-to-blood volume ratio, total reaction volume, concentration of sodium dodecyl sulfate, chamber design, were optimized order achieve quantitation capability across clinical range path length suitable for platform. Besides quantitative correlation with clinically accepted-validated standard method, spectral absorption characteristics hemoglobin–SLS mixture newly developed compared those assays. The finalized chip reagent, cost 0.136 USD. combination an automated microscope was able Pearson 0.99 validation study comparing commercially available hematology analyzer, turnaround time 10 min, incubation time. performance ascertained, sensitivity 92.3% specificity 53.8%.
